Define PHARMACOLOGY
The science of drug action on bioloical systems. Includes two disciplines: Pharmacokinetics and pharmacodynamics.
Define PHARMACOKINETICS
The quantitative study of:
Absorption, distribution, metabolism , and excretion
Define PHARMACODYNAMICS
The study of the effect (molecular, cellular, or biochemical) of drugs and their mechanism of action.
Define CLEARANCE
The volume of body fluid cleared of drug per unit time.
Define TOTAL BODY CLEARANCE
The sum of drug clearances by all routes of elimination (e.g. liver, kidney, lung, etc.)
Define VOLUME OF DISTRIBUTION
A proportionality constant that relates the dose of drug administration to the plasma concentration
Define ELIMINATION HALF LIFE
The time required for plasma drug concentration to decrease by 50%
Define BIOAVAILABILITY
F = Fraction of dose that reaches the systemic circulation following administration of any form
Define STEADY STATE PLASMA DRUG CONCENTRATION
CPss = The peak drug plasma concentration at steady state
Define MINIMUM TOXIC CONCENTRATION
The minimum drug plasma concentration which can elicit toxic effects
Define MINIMUM EFFECTIVE CONCENTRATION
The minimum drug plasma concentration needed to elicit its effect
Define THERAPEUTIC RANGE
A.K.A Therapeutic Window... the range of drug plasma levels between the minimum toxic concentration and minimum effective concentration
Define THERAPEUTIC INDEX
The minimum toxic concentration divided by the minimum effective concentration
What is the distribution phase?
Also known as the alpha phase, it is the portion of a plasma concentration-time curve after a single IV dose which represents movement of drug molecules across the membrane.
What is the elimination phase?
Also known as the beta phase, it is the portion of a plasma concentration-time curve after a single IV dose which represents gradual decrease in drug concentration due to metabolism and excretion.
