Why do we tilt a test tube when taking out liquid culture?
To prevent the culture from pouring out and minimizes the surface area of the tube exposed to contaminants in the air
How do we remove an inoculum form a slant?
Keep loop hand still, pull the tube away from the loop, the loop should scrape no more than 1/4 full of the bacteria
How do we lift the lid from a Petri dish?
Only about a 45-degree angle, NEVER lift the lid off completely to minmize contamination
