Define the following:
  • Pure culture
  • Colony
  • Lawn
  • Medium
  • TCS Plate
  • Pure Culture: Isolated group of genetically identical cells
  • Colony: Small mass of genetically identical cells (millions/billions) that can be viewed with the naked eye
  • Lawn: A dense and even layer of bacteria
  • Medium: Solution or solid that contains nutrients a cell can use
  • TCS Plate: Tripticase Soy Plate that contains enzymatically digested soy protein as a source of nitrogen
How do light microscopes work?
Light travels from the light source, shines on the specimen, and is then magnified by the lenses:
  • The condenser lens focuses the light on the specimen
  • The objective lens magnifies the image of the specimen
  • The ocular lens magnifies the image and inverts it for viewing
How do you calculate the total magnification of a microscope?
Magnification (ocular lens) x Magnification (objective lens0
What is Resolution?
The minimum distance required between two points for them to remain visibly identifiable as separate points. E.g. R = 0.3 micrometers means a microscope can resolve individual distances of those that are 0.3 micrometers or more apart
What is osmosis?
Movement of water across membranes from an area of low solute concentration (high solvent/water) to an area of high solute concentration (low solvent/water)
What is the scientific method?
  1. Formulate an idea/question
  2. Come up with a hypothesis (a good predication based on backgrroud knowledge and literature search)
  3. Perform experiment with experimental group (manipulated and responding variables) and control group
  4. Record results
  5. Based on results, use it to make a theory
Define the following:
  • Interpupillary distance
  • Ocular micrometer
  • Parfocal microscopes
  • Turgid
  • Plasmolyzed
  • Interpupillary distance: Distance between a person's pupils
  • Ocular Micrometer: Used to measure size of magnified objects
  • Parfocal Microscopes: Have objective lenses that can be changed without having to refocus the microscope
  • Turgid: Swollen
  • Plasmolyzed: The protoplast (contents of the cell minus the cell wall) contracts
What is the function of the cell wall?
  1. Constrains cytoplasm
  2. Prevents plasma membrane from rupturing when the cell enlarged due to water uptake
Describe what happens to a cell that is in:
  • Hypotonic
  • Hypertonic
  • Isotonic
conditions
Hypotonic: Water moves into the cell, the cell becomes turgidHypertonic: Water leaves the cell, the cell shrinks and becomes plasmolyzedIsotonic: No net movement of water, causing cell to become flaccid due to lack of pressure
What is a solvent control group?
The solvent control group confirms that the effect on the responding variable you are measuring is due only to the responding variable, and not the solvent it is dissolved in E.g., If you are testing the effect of chocoalte in chocolate milk in hyperactivity, you would need to also test the role of milk on hyperactivity
